人参皂苷Rg1配伍丹参酮IIA对缺氧-复氧H9c2细胞的保护作用及机制研究 被引量:9

摘要 目的研究人参皂苷Rg1配伍丹参酮IIA对缺氧-复氧H9c2细胞的保护作用及机制。方法培养H9c2细胞并分为空白对照组(DMEM处理、常规条件培养)、缺氧-复氧损伤组(DMEM处理、缺氧-复氧培养)、人参皂苷组(100μmol/L人参皂苷Rg1处理、缺氧-复氧培养)、丹参酮组(10μmol/L丹参酮IIA处理、缺氧-复氧培养)、联合用药组(人参皂苷Rg1和丹参酮IIA处理、缺氧-复氧培养),MTT法检测细胞活力、Western-blot方法检测Bcl-2、Bax的蛋白含量。结果人参皂苷组、丹参酮组、联合用药组的MTT值高于缺氧-复氧损伤组,且联合用药组的MTT值高于人参皂苷组、丹参酮组。联合用药组的Bcl-2含量高于人参皂苷组、丹参酮组,Bax含量低于人参皂苷组、丹参酮组。MTT值与Bcl-2含量呈正相关、与Bax呈负相关。结论人参皂苷Rg1配伍丹参酮IIA能够对缺氧-复氧H9c2细胞发挥保护作用,这一作用可能是通过上调Bcl-2、下调Bax来实现的。 [Objective ] To study the protective effect and mechanism of ginsenoside Rgl combined with tanshi- none IIA on H9c2 ceils treated by hypoxia-reoxygenation. [Methods] H9c2 ceils were cultured and divided into blank control group (treated by DMEM and rotttine conditions), hypoxia-reoxygenation injury group (treated by DMEM and hypoxia-reoxygenation), ginsenoside group (treated by 100 umol/L ginsenoside Rgl and hypoxia-reoxy- genation), tanshinone group (treated by 10 umol/L tanshinone IIA and hypoxia-reoxygenation), combined treatment group (treated by ginsenoside Rgl and tanshinone IIA and hypoxia-reoxygenation). Then cell viability was assayed by MTI" and protein levels of Bcl-2, Bax were detected by Western-blot. [ Results ] MTI" values of ginsenoside group, tanshinone group, combined treatment wele higher than hypoxia-reoxygenation injury group, and MTY values of combined treatment group were higher than ginsenoside group and tanshinone group. Bcl-2 levels of combined treatment were higher than ginsenoside group and tanshinone group, Bax levels were lower than ginsenoside group and tanshinone group. MTT values were positively correlated with Bcl-2 levels and negatively correlated with Bax levels. [ Conclusion ] Ginsenoside Rgl combined with tanshinone IIA can exert protective effects on hypoxia-reoxy- gcnation H9c2 cells and this effect might be completedby upregulation of Bcl-2 and downregulation of Bax.
