摘要
目的观察大鼠气道平滑肌细胞(ASMC)在不同浓度莫西沙星作用不同时间后,线粒体膜电位(△Ψm)及细胞增殖的变化,探讨莫西沙星抑制ASMC增殖的可能机制。方法体外原代培养大鼠ASMC,将传代培养的大鼠ASMC 5~6代于浓度为0、10、20、30和40μg/mL的莫西沙星中孵育不同时间(4、7和11 d)后,MTT法监测吸光值,计算细胞增殖抑制率;用JC-1荧光探针在免疫荧光显微镜下检测细胞△Ψm的变化。结果 10~40μg/mL莫西沙星作用于ASMC不同时间(4、7和11 d),明显抑制细胞增殖,并存在浓度(r=0.683,P〈0.01)及时间依赖性(r=0.568,P〈0.01);10~40μg/mL莫西沙星干扰ASMC△Ψm,同时存在浓度(r=-0.876,P〈0.01)及时间依赖性(r=-0.378,P〈0.05)。结论莫西沙星可能通过影响大鼠△Ψm以抑制ASMC增殖。
[Objective] To observe the effects of moxifloxacin at various concentrations and time on the alteration of △Ψm and the proliferation of ASMC,and to explore the possible mechanism. [Methods] ASMC was derived from rat airway tissue and cultured in vitro. It was randomly divided into five groups including a control group and four groups that moxifloxaein were added at different concentrations (10μg/mL,20μg/mL,30μg/mL,40μg/ mL). Then all the above groups were incubated for 4 d,7 d and 11 d. The absorbanee ratios of various groups were determined by MTT assay and the JC-1 probe was employed to measure mitochondrial depolarization in ASMC. [Results] Moxifloxacin at 10 -40μg/mL significantly inhibited ASMC' proliferation with concentration and time dependence (r =0.568, P 〈0.01; r =0.683, P 〈0.0l); With the increasing of the concentration of moxifloxaein and the extension of action time, there was a shift from mitoehondrial orange-red uorescenee to green uoreseenee among groups, also with a concentration and time dependence (r =-0.876, P 〈0.01; r =-0.378, P 〈0.05). [Conclusion] Moxifloxacin can inhibit ASMC' proliferation by altering △Ψm.
