摘要
目的检测血清同型半胱氨酸(homocysteine,Hcy)浓度及其相关胱硫醚β合成酶(cystathionineβsynthase,CBS)、甲硫氨酸合成酶(methionine synthase,MTR)在人体血清中RNA表达水平变化,探讨Hcy浓度、CBS、MTR表达水平与脑卒中的关系。方法收集242例脑卒中(脑出血118例、脑梗死124例)患者和78例健康人静脉血,用循环酶法进行Hcy含量测定,Real time-PCR法检测人血清中CBS、MTR相对RNA表达量。结果脑梗死组Hcy浓度(16.49±8.88)μmol/L、脑出血组Hcy浓度(15.48±9.78)μmol/L,高于正常对照组(8.29±2.68)μmol/L(P<0.05);正常对照组血清中CBS表达量高于脑卒中组,差异有统计学意义(P<0.05),正常对照组血清中MTR表达量高于脑卒中组,但差异无统计学意义(P>0.05)。脑梗死组与脑出血组间Hcy、CBS及MTR 3种指标比较差异无统计学意义(P>0.05)。结论脑卒中(脑出血、脑梗死)患者血清Hcy和CBS表达水平较正常人显著增高。
[Objective] To explore the relationship between the Hcy, CBS, MTR in patients with the stroke, detect the RNA express level of the serum homocysteine (Hcy), cystathionine β Synthase (CBS) and methionine Synthase (MTR). [Methods] Obtained blood from 242 patients with stroke (118 patients with in- tra-eerebral hemorrhage, 124 patients with brain infarction) and 78 healthy people.Detected the serum Hey concentration by enzymic method and the relative expression of CBS and MTR RNA in patients by Real time-PCR. [Results] The concentrations of serum Hey in brain infarction group (16.49 ± 8.88) μmol/L and intra-cerebral hemorrhage group (15.48 ± 9.78) μmol/L were higher than the control group (8.29 ±2.68) μmol/L (P〈0.05); the relative expression of CBS in healthy group was higher than that in the stroke group with statistical significance (P〈0.05); the relative expression of MTR in healthy group is higher than its in the stroke group without statistical significance (P〉0.05); the brain infarction group and intra-cerebral hemorrhage group showed no significant differences in these three indexes (P〉0.05). [Conclusion] Serum homocysteine and CBS have the positive significant value for diagnose stroke disease.
