摘要
目的探讨龙胆碱对发育期大鼠反复惊厥后海马神经元凋亡的影响。方法采用热水浴法制备大鼠惊厥模型,用TUNEL法测定海马区神经元凋亡情况。结果模型组较多神经细胞核呈棕褐色阳性反应,凋亡细胞面积显著增加,平均灰度明显增强;龙胆碱高、低剂量组细胞核棕褐色的阳性目标面积较模型组,平均灰度明显降低,与模型组比较有显著性差异,龙胆碱高、低剂量组之间差异无显著性。结论龙胆碱具有一定的抗凋亡作用,该作用可能是龙胆碱发挥抗惊厥性脑损伤作用的机制之一。
Objective To investigate the effect of Gentianine on apoptosis of hippocampus cells in a developing rat model of Febrile Convulsions( FC). Methods Warm water inducing rat FC model was developed in this study and the method of TUNEL was used to determine the apoptosis of hippocampus cell. Results Positive expression of hippocampus cells appeared brown and increased significantly in FC group,the average area of positive cells and positive target gray value increased markedly than that of control group. However,the area and target gray value in GT1 and GT2 groups decreased than that of FC group,and there was not difference in quality between the GT1 and GT2 group. Conclusion Gentianine had the role of inhibiting apoptosis of the hippocampus cells,which was the mechanism releasing and ameliorating brain injure caused by FC.
