TSA体外抑制人肺癌细胞系A549细胞增殖及机制

摘要 目的探讨TSA对人肺癌细胞系A549的作用及其对上皮间质转化(EMT)的调控机制。方法用MTT实验分析TSA体外抑制A549细胞的增殖能力;用划痕实验检测细胞的迁移;用Western blot法检测EMT相关marker蛋白的表达。结果 TSA对A549细胞的增殖具有明显的抑制作用(P<0.01),TSA通过EMT途径抑制A549细胞的侵袭与迁移。结论 TSA可体外抑制A549细胞,其部分机制是通过抑制EMT途径实现的。 Objective To investigate the effect of TSA on lung cancer cell A549 and its possible mechanism. Methods Proliferation of A549 cells was detected by MTY assay ; Migration of A549 cells was measured by Wound healing assay; The expression EMT markers proteins level were detected by Western blot. Results TSA signifi- cantly inhibited the proliferation of A549 cells( P 〈 0. 01 ). TSA inhibited A549 cells invasion and migration via in- hibition of EMT. Conclusions TSA may effectively inhibit the ability of proliferation and migration of lung cancer, and the mechnism may be explained by inhibition of EMT.
