罗格列酮抑制大鼠腹膜透析相关性腹膜纤维化中TGF-β_1和CTGF的表达 被引量:7

摘要 目的:观察罗格列酮(RGZ)对腹膜透析相关性腹膜纤维化中TGF-β1和CTGF的表达的影响。方法:36只SD大鼠随机分为4组:空白对照组、模型组、低浓度RGZ组、高浓度RGZ组(每组n=9)。除空白对照组外其余大鼠均置入腹透管,除空白组外其余组均采用高糖腹膜透析液+红霉素制作慢性腹膜透析腹膜纤维化模型。低浓度RGZ组、高浓度RGZ组分别采用1.5 mg/kg RGZ、15 mg/kg RGZ治疗。透析8周后处死大鼠,取壁层腹膜行H-E及Masson染色,观察大鼠腹膜形态;免疫组化检测腹膜转化生长因子(TGF-β1)和结缔组织生长因子(CTGF)的表达情况。结果:与空白对照组相比,模型组间皮下基质增厚,血管增生及炎症细胞浸润明显增加(P<0.05);TGF-β1、CTGF表达水平明显增高(P<0.05);RGZ能改善上述病理改变,下调TGF-β1、CTGF表达(P<0.05);两种剂量间差异无统计学意义(P>0.05)。结论:在大鼠腹膜透析相关性腹膜纤维化模型中加用罗格列酮能有效抑制TGF-β1及CTGF表达。 Objective:To investigate the effect of rosiglitazone( RGZ)in prevention of peritoneal fibrosis induced by perito-neal dialysis in rats. Methods:Thirty-six SD rats were randomly divided into four groups:control group(n=9),model group(n=9),low-dose RGZ group(n=9),high-dose RGZ group(n=9). Except for control group,peritoneal catheters were implanted in another Twenty-seven rats. At the end of the study. The visceral peritoneum tissues of rats were harvested and stained with hema-toxylin-eosin( H-E)and Masson trichrome. The expression of TGF-β1 and CTGF in parietal peritoneum were detected with im-munohistochemistry assay. Results:Compared with control group,the number of peritoneal vessels,leukocytes,peritoneal thickness and the expression of TGF-β1 and CTGF were significantly higher in model group group(P﹤0. 05). Administration of RGZ resulted in preserved the number of peritoneal vessels,leukocytes and peritoneal thickness,and reduced the expression of TGF-β1 and CTGF (P﹤0. 05). But there was no significant difference between the low-dose RGZ group and the high-dose RGZ group(P﹥0. 05). Conclusion:In rat of peritoneal dialysis-related peritoneal fibrosis,administration of rosiglitazone can inhibit the expression of TGF-β1 and CTGF effective.
