淫羊藿苷对AD细胞模型CREB表达的影响及机制研究 被引量:5

摘要 目的研究淫羊藿苷(ICA)对阿尔茨海默病细胞模型中CREB表达的影响及其作用机制。方法以Aβ25-35孵育PC12细胞制备的AD细胞模型为研究对象,分为5组,分别为对照组、Aβ组、ICA、LY组、ICA+LY组(LY为PI3K/Akt信号通路的经典抑制剂),Western-blot法测定各组p-CREB(Ser133)/CREB、p-Akt(Ser473)/Akt的蛋白表达水平。结果 Aβ组中P-CREB、P-Akt蛋白表达水平明显低于对照组(P<0.01),且ICA组中P-CREB、P-Akt蛋白表达水平明显高于Aβ组(P<0.01),ICA+LY组中P-CREB、P-Akt蛋白的表达水平明显低于ICA组(P<0.01)。结论 Aβ25-35能明显下调PC12细胞中P-CREB、P-Akt蛋白表达水平,而ICA干预后能上调上述蛋白的表达水平。ICA可能通过PI3K/Akt信号通路来上调P-CREB表达水平而发挥其保护作用。 Objective To explore the effects and mechanisms of Icariin on the expression of CREB in the Alzheimer's disease cell models.Methods AD cell models were divided into 5 groups,including Control group,Aβ group,ICA + Aβ group,Aβ + LY group,and ICA + Aβ + LY group(LY is the classical inhibitor of PI3K/Akt).The expressions of p CREB、CREB、p Akt and Akt in each cell group were detected by Western Blot.Results Compared with the control group,the level of P CREB and p Akt of the model group declined significantly.Compared with the Aβ group,the level of P CREB and p Akt of ICA group increased significanly (P<0.01).Compared with the ICA group,the level of P CREB and p Akt of ICA + LY group declined signif icantly (P<0.01).Conclusions Aβ25-35 can decrease the protein levels of P CREB and p Akt in PC12 cells; af ter ICA pretreatment,the protein level were significantly increased to protect cells.The protective effect of ICA on PC12 cells may relate with the increase expression of p CREB by the PI3K/Akt pathway.
