摘要
目的研究黄芩清热除痹组方对佐剂性关节炎大鼠血清中白细胞介素-8(IL-8)、白细胞介素-10(IL-10)、肿瘤坏死因子-α(TNF-α)表达及脏器指数的影响。方法 SD大鼠60只,雄性,随机分成6组,每组10只。除正常对照组外,采用弗氏完全佐剂诱导佐剂性关节炎(AA)大鼠模型。造模第12 d灌胃给予相应药物,1次/d,连续12 d。实验结束后,检测并比较各组大鼠脏器指数及血清中IL-8、IL-10、TNF-α含量。结果给药后,与正常组相比,模型组大鼠血清中IL-8、TNF-α含量显著性升高(P<0.01),IL-10含量及脏器指数显著降低(P<0.01)。与模型组比较,黄芩清热除痹组方组大鼠血清中IL-8、TNF-α含量明显降低(P<0.05,P<0.01),脏器指数及IL-10含量明显升高(P<0.05,P<0.01),其中高剂量组作用效果最明显(P<0.01)。黄芩清热除痹组方高剂量组与雷公藤多苷片对照组作用效果无显著性差异(P>0.05)。结论黄芩清热除痹组方可以降低AA大鼠血清中IL-8、1NF-α含量,升高脏器指数及血清中IL-10含量,从而发挥对AA大鼠抗炎消肿的作用。
Objective To study the influences of Huangqin Qingre Chubi Fang on content of interleukin8 (IL-8),interleukin-10 (IL-10),tumor necrosis factor-α (TNF-α) and visceral indexes in rats with adjuvant arthritis (AA).Methods Male SD rats (n =60) were randomly divided into 6 groups (each n =10).The rat model of AA was established with Freund' s complete adjuvant in all groups except of normal group.After modeling for 12 d,different groups were orally given corresponding medicinal once a day for 12 d.The visceral indexes and content of IL-8,IL-10 and TNF-α were detected and compared in all groups.Results After treatment,the content of IL-8 and TNF-α increased significantly (P 〈 0.01),and IL-10 content and visceral indexes decreased significantly (P 〈 0.01) in model group compared with normal group.The content of IL-8 and TNF-α decreased significantly (P 〈0.05,P 〈0.01),and IL-10 content and visceral indexes increased significantly (P 〈 0.05,P 〈 0.01) in Huangqin Qingre Chubi Fang group compared with model group,which,was more significant in high-dose group (P 〈0.01).The effective results had no significant difference between high-dose group and GTW Tablet group (P 〉 0.05).Conclusion Huangqin Qingre Chubi Fang can reduce the content of IL-8 and TNF-α and raise visceral indexes and the content of IL-10,and plays a role of anti-inflammation and detumescence in rats with AA.
