响应面法优化酶法去除龙芽楤木多糖中蛋白工艺的研究

Optimization of Deproteinized Process of Aralia elata Polysaccharide by Enzyme Method and Response Surface Methodology

摘要 确定木瓜蛋白酶去除龙芽楤木多糖中蛋白质的最佳工艺。在单因素试验基础上,采用Box-Behnken试验设计优化酶用量、时间、温度的最佳条件。酶法脱蛋白最优工艺条件为酶用量2.7%、时间29.4 min、温度52.0℃,蛋白质脱除率为57.4%。酶法脱蛋白效果较好,操作简捷,便于工业化生产。 Optimal technics of removing Aralia elata polysaccharide protein by papain was determined. Best conditions of papain dose, time and temperature were studided with single factor experiments and Box-Behnken experimental design. The results showed that the optimum conditions were as followings: papain dose 2.7%, time 29.4 min, temperature 52.0℃. Under these conditons, removing rate of protein was 57.4% and effect for deproteinization method was good, easy to operate and convenient for industrialized production.
