摘要
目的通过测定整合素β1 mRNA和蛋白的表达水平,研究硫酸右旋糖苷(DS)对人胃癌细胞(BGC-823)在裸鼠大网膜上种植转移的影响,从而观察DS预防胃癌细胞在腹腔种植转移中的作用。方法裸鼠共80只,实验组40只和对照组40只。实验组裸鼠腹腔内同时注入1×l07BGC-823悬液(0.2 mL·只-1)和0.3%的DS(1 mL·只-1);对照组腹腔内同时注入1×107胃癌细胞悬液(0.2 mL·只-1)和生理盐水(1 mL·只-1)。分别于注射后1、3、7、14 d处死,取大网膜组织,应用免疫组化、RT-PCR方法测定DS对整合素β1 mRNA和蛋白的影响。结果随着BGC-823在大网膜上种植时间的延长,DS实验组与对照组大网膜上瘤节数目增多且体积逐渐增大,实验组大网膜瘤结节数量与体积均显著低于对照组;免疫组化检测示,在不同时间点实验组细胞膜整合素β1的表达均显著高于对照组(P<0.05);在不同时间点上,实验组整合素β1 mRNA的表达均显著高于对照组(P<0.05)。结论 DS抑制胃癌细胞在裸鼠大网膜上的种植转移与整合素β1表达下调有关。
Objective To study the effect of dextran sulfate (DS) on the adhesion and metastasis of human gastric cancer cells in the abdominal cavity by investing the expression of the protein and mRNA of integrin β1. Methods Eighty - four BALB/c nude mice were divided into the experiment group (n = 40) and control group (n = 40) ;1 × 10 7 cancer cells in 0.2ml and 0.3% DS in l ml were peritoneally injected to each mouse in the experimental group;the same number of the cancer cells and 0.9% Nacl were peritoneally in- jected to each mouse in the control group. The mouse was sacrificed in the dl ,d3 ,d7 and d14 ,respectively;the omentum was harvested, The mass of the planting metastases of gastric cancer cells in the peritoneal cavity was observed by paunching, the expression levels of in- tegrin β1 mRNA were detected using the immunohistochemical analysis and reverse transcription - polymerase chain reaction ( RT - PCR). Results The size and the number of the tumor on the omentum increased along with the injecting time of the cancer cells in both the experimental group and the control group. The size of the tumor in the experimental was significantly smaller than that in the control group ; the number of the tumor in the experimental was significantly less than that in the control group. In the different time point, the ex- pression of the integrin 131 was significantly different, between the experimental group and control group, by immunohistochemical analysis ( P 〈 0.05 ) and RT - PCR ( P 〈 0.05 ). Conclusion DS inhibit the migration and invasion of gastric cancer cells in the abdominal cav- ity by downregulating the expression of integrin β1 mRNA to affect the integrin^l protein expression.
