神经生长因子对戊四氮致痫幼鼠海马细胞色素C表达的影响

Effects of Nerve Growth Factor on Expression of Cytochrome C in Hippocampus of Pentylenetetrazol-Induced Epileptic Young Rats

摘要 目的用戊四氮(PTZ)诱导建立的慢性癫痫(EP)幼鼠模型,研究神经生长因子(NGF)处理后幼鼠海马细胞色素C(Cyt C)的表达,探讨NGF在EP发病中的作用。方法 50只SD幼鼠随机分为阴性对照组(A组)、阳性对照组(B组)、NGF低剂量组(C组)、NGF中剂量组(D组)、NGF高剂量组(E组)。NGF用药后3d,取海马组织用于免疫组织化学及实时荧光定量PCR方法检测Cyt C的表达。结果与A组比较,B组Cyt C阳性细胞数及mRNA表达量明显增多(P<0.01)。与B组比较,C^E组Cyt C阳性细胞数及mRNA表达量降低(P<0.05)。结论 NGF可减少海马Cyt C的表达,促进EP幼鼠体质量增长。 Objective To explore the effects of nerve growth factor (NGF) on the expression of cytochrome c(Cyt C) in hippocampus of pentylenetetrazol (PTZ)-induced epileptic rats . Methods Fifty SD rats were randomly divided into 5 groups :negative control group (group A ) ,positive control group (group B) ,NGF low-dose group (group C) ,NGF medium-dose group (group D) and NGF high-dose group (group E) . After 3 days of NGF treatment ,the number of Cyt C positive cells and the expression of Cyt C mRNA in hippocampus were detected by immunohistochemisty and real-time PCR . Results The number of Cyt C positive cells and the expression of Cyt C mRNA in hippocampus of group B were significantly higher than group A (P〈0 .01) . The number of Cyt C positive cells and the expression of Cyt C mRNA of group C^E were lower than group B (P〈0 .05) . Conclusion NGF may play an impor-tant role in decreasing the expression of Cyt C and increasing the weight of epileptic rats .
