摘要 目的建立凝胶色谱-超高效液相色谱法(GPC-UPLC)测定食品中20种常见非食用色素的含量。方法食品样品经环己烷提取,离心后取上清液通过全自动凝胶色谱仪净化浓缩,净化液经0.45μm滤膜过滤,流动相乙腈-0.1%甲酸溶液,梯度洗脱,流速0.3 ml/min,采用Acquity UPLC BEH C18(50 mm×2.1 mm,1.7μm)柱分离,DAD检测器,检测波长210 nm^800 nm,以峰面积标准曲线法定量。结果 20种色素在15 min内实现完全分离,线性范围为1μg/ml^40μg/ml,相关系数0.879~1.000,检出限:0.01 mg/kg^0.3 mg/kg;回收率为60.3%~103.2%。结论该方法可靠、准确、灵敏度高,适合测定食品中的20种非食用色素的含量。 Objective To establish a gel permeation chromatography - ultra performance liquid chromatography (UPLC) meth- od for the determination of 20 Inedible pigments in foods. Methods 20 Inedible pigments were extracted with cyclohexane, and the supernatant were purified with GPC after centrifugation. Acetonitrile -0.1% formic acid as mobile phase for gradient e- lution at the flow rate of 0.3 ml/min, the separation was performed on UPLC BEH Cls (50 mm ×2.1 mm, 1.7 μm) column and detected with DAD at the wavelengths from 210 to 800nm. Results 20 Inedible pigments were separated well on BEH C18 column within 15 minutes,and the linear range is 1 μg/m1-40μg/ml The standard curve showed good linearity with the correlation coefficient of 0. 879 - 1. 000. The detection limits were 0.01 mg/kg -0.3 mg/kg and the recoveries were 60.3% - 103.2%. Conclusion The established method is reliable, accurate and sensitive and it could be applied for determination of 20 Inedible pigments in foods.
