摘要
目的探讨绒毛和蜕膜组织中氧化应激反应因子人过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPX1)、过氧化物歧化酶1(SOD1)、过氧化物歧化酶2(SOD2)、硫氧还蛋白1(TXNL1)、氧化型低密度脂蛋白受体1(LOX-1)与反复自然流产的关系。方法选取浙江大学医学院附属妇产科医院,2010年-2011年收治的46例反复自然流产患者、50例正常早孕妇女绒毛和蜕膜组织,应用实时荧光RT-PCR方法检测6种参与氧化应激反应的因子CAT、GPX1、SOD1、SOD2、TXNL1和LOX-1 mRNA的表达。结果早孕绒毛、蜕膜组织中均有氧化应激反应因子CAT、GPX1、SOD1、SOD2、TXNL1和LOX-1 mRNA的表达;前5种在绒毛、蜕膜组织中的相对表达量自然流产组均与对照组差异无统计学意义(P>0.05),氧化型低密度脂蛋白受体1(LOX-1)在自然流产组蜕膜和绒毛组织中高表达,与对照组比较差异有统计学意义(P<0.05)。结论氧化应激反应因子表达于早孕母胎界面绒毛和蜕膜组织中,绒毛、蜕膜组织中LOX-1mRNA的差异表达可能与早期自然流产发生机制相关。
Objective To explore the correlations between catalase ( CAT), Glutathione peroxidase 1 ( GPX1 ), superoxide dismutase 1 ( SOD1 ), superoxide dismutase 2 ( SOD2), Thioredoxin - like protein 1 ( TXNL1 ), lectin - type oxidized LDL receptor ( LOX - 1 ) and recurrent spontaneous abortion. Methods The villus and decidua tissues of recurrent spontaneous abortion groups (n = 46), and normal early pregnancy women (control group, n = 50) were collected at our hospital from 2010 to 2011. And their expressions of CAT, GPX1, SOD1, SOD2, TXNL1 and LOX - 1 mRNA were determined by realtime fluorescence quantitative PCR(polymerase chain reaction). Results The CAT, GPX1, SOD1, SOD2, TXNL1 and LOX- 1 mRNA gene expressions were detected in first trimester placental villus and decidua tissues. And first five gene expressions of mRNA in villus and decidua tissues in spontaneous abortion and control group showed no significantly differences (P 〉 0.01 ), except that the gene expressions of LOX - 1 mRNA in decidua and villus tissues were significantly higher than that in the control group (P 〈 0.05). Conclusion CAT, GPX1, SOD1, SOD2, TXNL1 and LOX - 1 are co - expressed in human villus and decidua tissues. The different LOX - 1 mRNA expression may play important roles in the pathogenesis of early spontaneous abortion.
