偶氮氯磷Ⅲ-Zn(Ⅱ)共振散射光谱法测定脑脊液和人血清中总蛋白 被引量:1

Determination of total protein in cerebrospinal fluid and human serum by chlorophosphonazo Ⅲ- Zn(Ⅱ) resonance light scattering method

摘要 目的建立简单、灵敏的测定脑脊液和人血清中总蛋白的新方法。方法在pH 4.0的BR缓冲溶液中,往偶氮氯磷Ⅲ-Zn(Ⅱ)体系中加入蛋白质,体系的共振散射信号线形增加,根据信号增加值计算蛋白质含量。结果该法的线性范围为0 mg/L^2.6 mg/L,检出限为29.3 ng/L,回收率为95%~102%,批内变异系数(CV)和批间变异系数分别为2.4%、2.9%。将该方法用于人血清和脑脊液中蛋白质的测定,与医院测定结果基本一致。结论本方法简便、灵敏,具有临床应用价值。 Objective To establish a simple and sensitive method for determination of total protein in cerebrospinal fluid and human serqm samples. Methods In pH 4.0 Britton - Robinson buffer, the adding of protein could lead to great enhancement of the resonance ]ight scattering signal (RLS) of Chlorophosphonazo Ill - Zn( I1 ). The concentration of protein was calculated according to the value of RLS enhancement. Results The method was linear in the range of 0 mg/L - 2.6 mg/L, the detection limit was 29.3 ng/L, the recovery was in the range of 95% - 102% , and the within - run and between - run CV were 2.4% and 2.9%, respectively. The method has been applied to the determination of total protein in cerebrospinal fluid and human serum samples, and the results were in agreement with those reported by the hospital. Conclusion This method is simple, sensitive and suitable for clinical application.
