摘要
以含70%EGCG的茶多酚为原料,单分散15μm的聚苯乙烯-二乙烯基苯微球为分离介质,以安全性较好的乙醇-水为洗脱剂,结合回收率和产率为考察对象,从流动相比例、上样量以及流动相流速3个方面来探索最佳的制备色谱纯化EGCG的工艺条件。在流动相为乙醇/水=30/70(v/v)、上样量为900 mg、流速为10 mL/min的条件下,分离得到EGCG的单体纯度为97.49%,回收率75.81%。该方法具有分离效率高、处理量大、产品安全性好、生产周期短等特点,为高纯度EGCG的工业化生产提供了一种有效的技术支持。
Abstract: In this paper, containing 70% EGCG of the tea polyphenols was used as raw material, 15 μm monodisperse polystyrene - divinylbenzene microspheres was used as a separation medium, with better security ethanol-water was used as eluent. Combined recovery and yield as the object, from the ratio of the mobile phase, the sample volume and flow rate of the mobile phase to explore the optimal conditions for preparing chromatographic purification of EGCG. Under the proportion of mobile phase (ethanol/water) was 30/70 (v/v), the sample volume was 900 mg, and the flow rate 10 mL/min operating conditions, the obtained EGCG purity was 97.49%, and the recovery rate achieved 75.81%. This method has superiority of large capacity, highly efficient, product safety and short production cycle, providing a favorable reference for the industrial production of high purity EGCG.
