清热解毒经验方对子宫内膜癌移植瘤的作用及TAMs表达的影响 被引量:7

Effect on Xenograft Tumor of Endometrial Carcinoma Treated with Empirical Formula for Clearing Heat and Detoxification and the Impacts on TAMs Expression

摘要 目的探讨清热解毒经验方对子宫内膜癌移植瘤的作用及对肿瘤相关巨噬细胞(TAMs)表达的影响。方法对子宫内膜癌细胞进行传代培养,建立裸鼠子宫内膜癌移植瘤模型,采用免疫组织化学染色的方法研究子宫内膜癌裸鼠移植瘤组织中TAMs的表达。结果孕激素组及成瘤前中药组、成瘤后中药组肿瘤生长速度均明显低于空白对照组(P<0.05),孕激素组及成瘤前中药组、成瘤后中药组肿瘤生长速度比较,差异无统计学意义(P>0.05)。TAMs在肿瘤组织中的表达,孕激素组、成瘤前中药组、成瘤后中药组与空白对照组比较明显降低(P<0.05);孕激素组与成瘤前中药组、成瘤后中药组比较明显降低。(P<0.05)。结论清热解毒经验方具有抑制肿瘤生长的作用,其可能是通过抑制TAMs在肿瘤组织中的表达而发挥抑制肿瘤生长的作用。 Objective To discuss the effect on xenograft tumor of endometrial carcinoma treated with empirical formula for clearing heat and detoxification and the impacts on tumor-associated macrophage (TAMs) expression.Methods The cells of endometrial carcinoma were subcultured to set up xenografi tumor of endometrial carcinoma in nude mice.IHC staining was adopted to study TAMs expression in xenograft tumor tissue of endometrial carcinoma.Results The tumor growth speed in the progestogen group,the Chinese medicine before tumor formed group and the Chinese medicine after tumor formed group was apparently lower than that in the blank control group(P <0.05).The difference was not significant in comparison among the progestogen group,the Chinese medicine before tumor formed group and the Chinese medicine after tumor formed group(P > 0.05).TAMs expression in the progestogen group,Chinese medicine before tumor formed group and Chinese medicine after tumor formed group was significantly reduced as compared with the blank control group(P < 0.05).That in the progestogen group was reduced apparently as compared with the Chinese medicine before tumor formed group and the Chinese medicine after tumor formed group(P < 0.05).Conclusion Empirical formula for clearing heat and detoxification acts on the inhibition of tumor growth possibly through suppressing TAMs expression in tumor tissue.
