摘要
目的 探讨NF-κB和AQP-3在退变椎间盘中的表达,并进一步探讨NF-κB和AQP-3在椎间盘退变中表达的相关性,为研究和治疗椎间盘退变疾病提供新的策略。方法 根据术前患者MRI资料观察的椎间盘退变情况,按照Videman分级分标准将标本分为三组:A组评分为1-3分(轻度椎间盘退变组),B组评分为4-6分(中度椎间盘退变组),C组评分为7~9分(重度椎间盘退变组)。应用免疫组织化学染色检测腰椎间盘髓核NF-κB及AQP-3的表达变化,并探讨NF-κB和AQP-3在椎间盘退变过程中的相互作用。结果随着椎间盘退变的加重,NF-κB的表达逐渐升高(〈0.01),而AQP-3的表达逐渐降低(〈0.01),在椎间盘退变加重过程中,NF-κB和AQP-3之间的表达呈负相关(=-0.805,〈0.01)。结论 NF-κB及AQP-3在退变的椎间盘中均有表达,且NF-κB和AQP-3的表达存在明显的负相关性。
【Objective】To detect the expressions of nuclear factor kappa B(NF-κB) and aquaporin-3(AQP-3)in human degenerated lumbar discs and explore the roles of NF-κB and AQP-3 in the process of intervertebral disc degeneration. 【Methods】30 degenerated lumbar intervertebal disc tissues were divided into three groups: group A score 1 to 3 points(mild intervertebral disc degeneration group); group B score 4 to 6 points(moderate intervertebral disc degeneration group); group C score 7 to 9 points(severe intervertebral disc degeneration group). The expressions of NF-κB and AQP-3 in degenerated lumbar discs were detected by immunohistochemistry. And the expression of NF-κB and AQP-3 were analyzed by Image proplus 6.0 image analysis software. 【Results】The expression of NF-κB was gradually increased with the aggravation of intervertebral disc degeneration, while AQP-3 was gradually reduced(P〈0.01). Moreover, the expression of NF-κB and AQP-3 was negatively correlated in the process(r =-0.805,P〈0.01). 【Conclusions】Both NF-κB and AQP-3 were detected in the degenerated intervertebral disc. There exist clear negative correlation between expression of NF-κB and AQP-3.
