摘要
目的开发一种可以快速、准确检测肺炎衣原体的检测方法。方法采用环介导等温核酸扩增技术(LAMP)对肺炎衣原体的特异性DNA保守片段进行扩增,将扩增的DNA与基因芯片进行杂交,观察是否可在芯片相应的区域出现杂交信号。结果只有肺炎衣原体DNA经LAMP反应后出现特异性的扩增条带,经LAMP扩增后的产物与基因芯片杂交,仅肺炎衣原体区域出现杂交信号。结论 LAMP技术联合基因芯片技术具有高度敏感性、高度特异性的特点,可快速准确地检测肺炎衣原体。
Objective To invent a rapid and accurate method to detect Chlamydia pneumonia (CP). Methods The specific DNA conservative segment of CP was amplified by using loop-mediated isothermal amplification(LAMP).The amplified DNA and gene chip were hybridized.Whether the hybridization signal appearing or not in the corresponding region of the chip was observed. Results Only the amplified CP-DNA by LAMP appeared the specific amplification bands.After the LAMP sequence was hybridized with gene chip,only in the CP area appeared corresponding hybridization signal. Conclusion LAMP combined with gene chip technology have characteristics of high sensitivity,high specificity,can detect CP quickly and accurately.
