麝香乌龙丸对佐剂关节炎大鼠垂体组织形态及ACTH、β-EP表达的影响 被引量:4

Effects of Shexiang Wulong Pills on the Tissue Morphology of Pituitary Gland and Expressions of ACTH and β-EP in Adjuvant Arthritis Rats

摘要 目的探讨麝香乌龙丸对类风湿关节炎(RA)大鼠的镇痛作用及可能机制。方法将60只Wistar大鼠随机分为正常组、模型组和麝香乌龙丸低、中、高剂量组,每组各12只。除正常组外其余大鼠均注射完全弗氏佐剂造模,于造模第15天起,麝香乌龙丸低、中、高剂量组分别给予麝香乌龙丸0.3、0.6、1.2 g/(kg·d),正常组与模型组灌胃等容量生理盐水,每日1次,连续给药2周。2周后处死取材,HE染色后在光镜下观察垂体组织形态,免疫组化法检测垂体促肾上腺皮质激素(ACTH)、β-内啡肽(β-EP)的表达。结果光镜下观察各组垂体组织形态均无明显改变。模型组和麝香乌龙丸低、中、高剂量组ACTH与β-EP表达均明显高于正常组(P<0.01)。麝香乌龙丸中、高剂量组ACTH表达明显低于模型组和麝香乌龙丸低剂量组(P<0.05或P<0.01)。麝香乌龙丸低、中、高剂量组β-EP表达均明显高于模型组(P<0.01)。麝香乌龙丸中、高剂量组β-EP表达均明显高于麝香乌龙丸低剂量组(P<0.05或P<0.01)。结论麝香乌龙丸可调节RA大鼠垂体ACTH、β-EP的表达,这可能是该药治疗RA疼痛的机制之一。 Objective To research the analgesic effect and possible mechanism of Shexiang Wulong Pills for rheumatoid arthritis( RA). Methods Sixty Wistar rats were randomized into the normal group,model group and lowdose,middle-dose and high-dose Shexiang Wulong Pills groups,with 12 in each. All rats were injected with complete Freund's adjuvant for modeling except rats in the normal group. From the 15 th day of modeling,the low-dose,middledose and high-dose Shexiang Wulong Pills groups were gavaged with 0. 3,0. 6 and 1. 2 g /( kg·d) Shexiang Wulong Pills once daily for 2 weeks. The normal group and model group were gavaged with equal volume of normal saline once daily for 2 weeks. The rats were sacrificed 2 weeks later. The tissue morphology of pituitary was observed under light microscope after HE staining. The expressions of adrenocorticotropic hormone( ACTH) and β-endorphin( β-EP) in the pituitary were detected with immunohistochemistry. Results The tissue morphology of pituitary was not significantly changed. Comparing with the normal group,the expressions of ACTH and β-EP were significantly increased in the model group and low-dose,middle-dose and high-dose Shexiang Wulong Pills groups( P〈0. 01). Comparing with the model group and low-dose Shexiang Wulong Pills group,the ACTH expression was significantly decreased in the middledose and high-dose Shexiang Wulong Pills groups( P〈0. 05 or P〈0. 01). Comparing with the model group,the β-EP expression was significantly increased in the low-dose,middle-dose and high-dose Shexiang Wulong Pills groups( P〈0. 01). Comparing with the low-dose Shexiang Wulong Pills group,the β-EP expression was significantly increased in the middle-dose and high-dose Shexiang Wulong Pills groups( P〈0. 05 or P〈0. 01). Conclusion Shexiang Wulong Pills can regulate the expressions of ACTH and β-EP in the pituitary of RA rats,which may be one of the mechanisms of analgesia.
