黄连解毒汤对热毒所致兔动脉粥样硬化及内皮祖细胞的影响 被引量:10

Effects of Huanglian Jiedu Decoction on the rabbit atherosclerosis caused by toxic heat and endothelial progenitor cells

摘要 目的通过建立热毒所致的兔动脉粥样硬化(Atherosclerosis,AS)模型,研究黄连解毒汤(Huanglian Jiedu Decoction,HJD)对兔动脉粥样硬化及内皮祖细胞(Endothelial progenitor cells,EPCs)的影响。方法在相同实验条件下,随机将30只新西兰雄兔分为5组,各组均静脉注射LPS(Lipopolysaccharides,LPS)联合高脂饲料喂养建立动脉粥样硬化模型,药物干预治疗前后,分别测定兔血脂的变化情况;抽取兔骨髓进行单个核细胞诱导培养、鉴定,加入含药血清后进行内皮祖细胞功能测定;观察腹主动脉及心脏组织病理形态学改变,了解AS形成情况及药物干预效果。结果建模4周末兔血脂水平达到高脂状态,中、高剂量黄连解毒汤及通心络干预组8周末血脂水平较4周末明显下降;黄连解毒汤中、高剂量组、通心络组含药血清对内皮祖细胞功能有明显促进作用;病理形态学观察,黄连解毒汤中、高剂量组、通心络组有显著减轻动脉粥样硬化形成的程度。结论黄连解毒汤可能通过降低血脂水平、抑制炎症反应及增强内皮祖细胞功能来延缓或防止热毒所致动脉粥样硬化的形成,为临床动脉粥样硬化的治疗提供参考。 Objective To establish an atherosclerosis model which is caused by toxic heat and study effects of Huanglian jiedu decoction( HJD) on the rabbit atherosclerosis and endothelial progenitor cells. Methods Under the same experimental conditions,30 New Zealand male rabbits were divided into 5 groups at random,all groups are intravenously injected LPS through the rabbit ear vein,meanwhile feeding the same high-fat diet as to establishing the atherosclerosis model. It's to test rabbit blood fats after4 weeks and 8 weeks for understanding the changed situation. Mononuclear cells of rabbit were isolated by bone marrow puncture and were cultured in vitro. When the cells passage culture is to reach P3,it should be appraised. After treatment,the EPCs of all groups should be determined cellular functions until P4. It is to observe changes of the hematoxylin-eosin( H-E) staining abdominal aorta and heart tissue pathological morphological with the optical microscope,understand the formation states of atherosclerosis and the effects of drug treatment. Results At the 4th weekend rabbit blood lipid level has achieved high fat state and it is significantly lower in middle-dose HJD treatment,high-dose HJD treatment and Tongxinluo treatment group at the 8th weekend. Compared with model group,medicated serum of middle-dose HJD treatment,high-dose HJD treatment and Tongxinluo treatment groups have obviously promoting effects of EPCs functions. For the pathological morphological observation,middle-dose HJD treatment,high-dose HJD treatment and Tongxinluo treatment groups significantly alleviated the degree of formation of atherosclerosis. Conclusion HJD could delay or prevent the formation process of atherosclerosis which is caused by toxic heat through lowering blood lipid level,inhibiting the inflammatory response and strengthen the functions of EPCs,it will provide reference for clinical treatment of atherosclerosis.
