射干麻黄汤加味对哮喘小鼠TGF-β_1的影响 被引量:7

摘要 目的:探讨射干麻黄汤加味对慢性哮喘小鼠TGF-β1的影响。方法:健康清洁级雌性BALB/c小鼠50只,每组10只,随机分为A组:正常对照组;B组:哮喘模型组;C组:地塞米松组;D组:射干麻黄汤组;E组:射干麻黄汤加味组。建立慢性哮喘小鼠模型后,分别予以相应药物干预,取材后分别采用ELISA法、免疫组化、实时定量PCR技术等方法检测BALF中TGF-β1含量、肺组织TGF-β1及TGF-β1mRNA表达强度的变化情况。结果:与正常对照组相比哮喘小鼠的BALF中TGF-β1含量、肺组织TGF-β1及TGF-β1mRNA表达强度均明显增加(P<0.05);经给予地塞米松、中药处理后上述指标均明显降低,哮喘发作程度减轻(P<0.05),其中射干麻黄汤加味组优于射干麻黄汤组(P<0.05)。结论:射干麻黄汤加味可抑制BALB/c哮喘小鼠哮喘的发生,其机制有可能是通过降低TGF-β1的表达而实现的。 Objective: To investigate the effect of modified She-gan-ma-huang decoction on chronic asthmatic mice of TGF-β1. Methods : Healthy clean grade female 50 BALB/c mice, 10 mice in each group, were randomly divided into group A : normal control group; group B : asthma model group; group C : dexamethasone group; group D: She-gan-ma-huang decoction group; group E : modified She-gan-ma-huang decoction group. To establish the model of chronic asthma mice, were given corresponding drug intervention, derived by ELISA method, immunohistochemistry, real-time quantitative PCR technique to detect BALF in lung tissue of TGF- β1 levels, TGF- β1 and TGF- β1 mRNA expression intensity. Results: Compared with the normal control group, asthmatic micse BALF TGF- β1 levels, lung tissue of TGF- β1 and TGF- β1 mRNA expression intensity was significantly increased (P 〈0. 05) ; the administration of dexamethasone, traditional Chinese medicine treatment of above indexes were significantly decreased, the degree of asthma attack also ( P 〈 0. 05 ), the modified She-gan-ma-huang decoction group is better than She-gan-ma-huang decoction group ( P 〈 0. 05) . Condusion: Modified She-gan-ma-huang decoction can inhibit the BALB/c of asthmatic mice with asthma, its mechanism may be achieved through reducing the expression of TGF- β1.
