摘要
探讨羊栖菜多糖、姬松茸多糖对高糖培养的胰岛β细胞的保护作用及其机制。将胰岛β细胞随机分为5组:正常对照组、模型组、SFPS组、ABP组、两药联合组。应用MTT法检测对胰岛β细胞增殖能力的影响,应用荧光显微镜检测胰岛β细胞凋亡情况,并用RT-PCR法检测bax、caspase-3基因表达量的变化。结果表明,SFPS、ABP能明显减轻高糖对胰岛βTc3细胞的生长抑制,抑制高糖诱导的胰岛β细胞凋亡,并明显降低bax、caspase-3基因表达(p<0.01),两药联合作用更明显,其作用机制可能与调控bax、caspase-3基因表达有关。
The protective effect and mechanism of SFPS and ABP on pancreatic beta cells in high glucose were studied. Pancre- atic βTc3 ceUs were randomly divided into model group, SFPS group, ABP group, SFPS + ABP group, and normal control group. The cells proliferation by MTF and cell apoptosis of βTc3 cells by fluorescence microscope were observed. The levels of caspase - 3 gene expression in all groups by RT-PCR were detected . SFPS and ABP were inhibiting proliferation inhibition rate of βTc3 cells, and reducing apoptosis of βTc3 cells. SFFS and ABP could effectively decrease the apoptotic rate of βTc3 cells and the level of bax and caspase - 3 gene expression. And the effect was more obviously in SFPS + ABP, it could effectively de- crease the apoptotic rate of βTc3 ceils probably through regulating bax and caspase -3 gene expression.
