姜黄素对纳米级二氧化硅粉尘所致大鼠肺损伤的抗氧化作用研究 被引量:2

摘要 目的:研究姜黄素对纳米级二氧化硅(nm-SiO2)粉尘诱导的大鼠肺损伤的抗氧化作用。方法:Wistar雄性大鼠50只,随机分为阴性对照组、模型组及姜黄素干预高、中、低剂量组,每组10只。气管内1次性灌注纳米级二氧化硅悬液(15 mg/100 g)复制大鼠肺损伤模型,姜黄素分别按200、100、50 mg/kg·d于大鼠造模后第2 d灌胃,4周后取动物血、肝、肺组织测定氧化损伤,另取一侧肺组织制作病理检查。结果:模型组大鼠体重低于阴性对照组(P<0.05),肺脏系数高于阴性对照组(P<0.01)。姜黄素各剂量组大鼠体重均高于模型组(P<0.05);与阴性对照组比较,模型组血清、肺组织匀浆、肝组织匀浆中超氧化物歧化酶(superoxide dismutase,SOD)活性、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)活性和总抗氧化能力(total-antioxide capacity,T-AOC)明显降低(P<0.01),丙二醛(malondialdehyde,MDA)含量增高(P<0.05);与模型组比较,血清、肺组织匀浆、肝组织匀浆中MDA含量降低;各剂量组两两比较,肺组织匀浆中MDA含量差异有统计学意义(P<0.05);随着姜黄素剂量的增加,呈一定的剂量反应关系。其中血清、肺组织匀浆、肝组织匀浆中SOD活性、GSH-Px活性及T-AOC含量呈现增高趋势,MDA含量呈现下降趋势。结论:姜黄素能对抗大鼠氧化损伤的过程,缓解由纳米二氧化硅粉尘引起的氧化损伤。 Objective:To explore the antioxidative effect of curcumin on the injured lung of the rats induced by nm - sized silicon dioxide(SiO2 )dust. Methods:50 male Wistar rats were randomized into 5 groups,namely,negative contrast group, model group and three curcumin intervention groups(high dosage,medium dosage and low dosage),with 10 rats in each group. Rats in both the model group and the three curcumin intervention groups were once given gastro - intillation of nm - sized SiO2 suspension(15 mg/ 100 g)through trachea to establish rat lung injury models. The rats from three curcumin intervention groups were given gastro - intillation of curcumin,with doses of 200,100 and 50 mg/ kg·d,two days after the establishment of the models. The tissues of hearts,blood,lung and the liver of the rats were taken to examine oxidative injury,with the tissues of the other side of the lung taken for pathological section. Results:The body weight of the rats in the model group was significantly lower than that in the negative contrast group(P ﹤ 0. 05),with the co - efficient of heart and lung higher than that in the nega-tive contrast group(P ﹤ 0. 01). The body weight of the rats in the three curcumin intervention groups was higher than that in the model group(P ﹤ 0. 05). Compared with that of those in the negative contrast group,the activity of superoxide dismutase (SOD),glutathione peroxidase(GSH - Px)and total - antioxide capacity(T - AOC)in serum,lung,and liver homogenate of the rats from the model group decreased significantly(P ﹤ 0. 01),with the content of malondialdehyde(MDA)significantly in-creased(P ﹤ 0. 05). Compared with that in the model group,MDA content decreased in the serum,lung,and the liver homog-enate of the rats from the three curcumin intervention groups;there was significant difference in MDA content among the three curcumin intervention groups(P ﹤ 0. 05),indicating a dosage - effect causal relation with increase in the dose of curcumin;the activity of SOD,GSH - Px and T - AOC increased in the serum,lung,and liver homogenate. Conclusion:Curcumin can fight off the process of oxidative damage in rats and alleviate the oxidative damage caused by nm - sized SiO2 .
