泰山磐石散对多囊卵巢综合征大鼠血清肿瘤坏死因子α表达及妊娠结局的影响 被引量:3

摘要 目的观察泰山磐石散对多囊卵巢综合征(PCOS)大鼠血清肿瘤坏死因子α(TNF-α)表达及妊娠结局的影响,探讨TNF-α与妊娠的关系。方法采用脱氢表雄酮建立PCOS大鼠模型70只,另取20只正常雌鼠,与雄鼠按1∶1合笼使其受孕,分为正常妊娠组、PCOS妊娠空白组、PCOS妊娠中药组及PCOS妊娠西药组,每组各20只。妊娠第1 d开始,PCOS中药组予泰山磐石散颗粒溶液1 mL/100 g体质量灌胃给药,PCOS妊娠西药组予等容积地屈孕酮溶液灌胃,PCOS妊娠空白组予等容积0.9%氯化钠注射液灌胃。均每日1次,连续5 d。正常妊娠组不进行任何干预。以上各组孕鼠均于妊娠第5 d取6只,眼眶静脉采血,进行血清TNF-α的测定。妊娠第8 d另取各组孕鼠6只,脱臼处死,取双侧子宫,记录孕鼠数及胚胎着床数。结果PCOS妊娠空白组血清TNF-α水平较正常妊娠组明显升高(P<0.05),胚胎着床平均数降低(P<0.05);而PCOS妊娠中药组及PCOS妊娠西药组血清TNF-α较PCOS妊娠空白组降低(P<0.05),胚胎着床平均数升高(P<0.05),与正常妊娠组比较差异无统计学意义(P>0.05)。结论 PCOS妊娠大鼠血清TNF-α水平较正常妊娠大鼠偏高,泰山磐石散能在一定程度上降低血清TNF-α的水平,并能提高胚胎着床率。血清中TNF-α浓度水平与妊娠结局存在一定相关性,有望成为预测流产的指标之一。 Objective To study the effect of Taishanpanshi powder on tumor necrosis factor (TNF - α) expression of poIycystic ovary syndrome (PCOS) rats serum and the pregnancy outcome, and explore the correlativity between serum TNF -α and pregnancy. Methods 70 PCOS rat models were established by Dehydroepiandros- terone. 20 normal male rats were selected with male mice according to 1 : 1 cage make its conception. Then these rats were divided into four groups, normal pregnant control group, PCOS pregnancy control group, the traditional Chinese medicine group and Western medicine group. PCOS traditional Chinese medicine group received aishanpanshi powder solution, 1 mL/100 g (body weight). PCOS Western medicine group received dydrogesterone isometric solution. 20 P- COS female rats in PCOS pregnancy control group received 0.9% sodium chloride injection to fill the stomach. Normal pregnancy group was not given any intervention. Pregnant rats were in pregnant 5 d, orbital venous blood, to determine the TNF alpha. In pregnant 8 d, from bilateral uterine recorded the number of blastocyst implantation and pregnancy rats. Results Serum TNF - α level in PCOS pregnancy control group was higher than that in normal pregnant control group ( P 〈 0.05 ), the number of blastocyst implantation was reduced ( P 〈 0.05 ). TNF - ot level in the traditional Chinese medicine group and Western medicine group were increased compared with that in PCOS pregnancy control group (P 〈 0.05), and the high number of blastocyst implantation. There was no significant difference be- tween the traditional Chinese medicine group and Western medicine group and normal pregnant control group ( P 〉 0. 05 ). Conclusion TNF - α level in PCOS pregnancy rat serum was higher than that in normal pregnancy, Taishanpanshi powder can decrease TNF - α level in pregnancy rat serum and increase the number of blastocyst implantation. Serum TNF - α levels have a certain relationship with pregnancy outcome, is expected to become one of the indexes prediction abortion.
