摘要
目的:探讨葡萄糖调节蛋白78(glucose regulated protein,GRP78)与细胞衰老在宫颈癌发生、发展过程中的相关性。方法:免疫组化法检测宫颈癌组织芯片中衰老相关异染色质灶标志蛋白HP1-γ及GRP78蛋白表达情况。结果:正常宫颈组织中HP1-γ蛋白及GRP78蛋白均呈低表达,而在宫颈上皮内瘤变(cervical intraepithelial neoplasia,CIN)Ⅱ-Ⅲ组织中HP1-γ蛋白及GRP78蛋白表达均逐渐升高。随着宫颈癌恶性程度的升高,GRP78表达明显升高,呈强阳性表达,而HP1-γ蛋白表达逐渐降低至较低水平。结论:GRP78蛋白可能通过抑制细胞衰老从而导致宫颈细胞由正常向恶性转化。
Objective: To explore the correlation between glucose regulated protein( GRP78) and cell senescence in occurrence and development of cervical cancer. Methods: Immunohistochemical method was used to detect expressions of senescence- associated heterochromatin foci HP1- γ and GRP78 protein in cervical cancer tissue. Results: The expressions of HP1- γ protein and GRP78 protein in normal cervical tissue were low,which increased gradually in cervical intraepithelial neoplasia( CIN) Ⅱ- Ⅲ. GRP78 expression increased significantly with aggravation of cervical cancer,which was positive strongly,but HP1- γ protein expression decreased to a lower level.Conclusion: GRP78 protein may induce malignant transformation of cervical cells through inhibiting cell senescence.
