基于牛黄特征峰的安宫牛黄丸特征指纹图谱研究 被引量:9

Study on characteristic fingerprints of Angong Niuhuang Pill based on the characteristic peaks of Bovis Calculus

摘要 目的:建立安宫牛黄丸特征指纹图谱的方法,以鉴别含有不同品种牛黄的安宫牛黄丸制剂。方法:以水饱和二氯甲烷-甲醇-冰醋酸(100:50:2)为提取溶剂,溶剂挥干,甲醇转溶后采用C18反相色谱柱分离,流动相为乙腈-0.5%磷酸水梯度洗脱,检测波长200nm。结果:通过对提取溶剂、检测波长及系统方法学考察,建立了不同品种牛黄及其制剂安宫牛黄丸特征指纹图谱,并确定了安宫牛黄丸特征指纹图谱中天然牛黄的特征峰。结果表明,通过该特征图谱的分析可以鉴别天然牛黄和培植牛黄以及含有天然牛黄和培植牛黄的安宫牛黄丸,该方法稳定性和重现性良好。结论:此方法能够反映安宫牛黄丸中天然牛黄的特征,鉴别含有天然牛黄和培植牛黄的安宫牛黄丸制剂,可用于安宫牛黄丸的整体质量评价。 Objective: To establish the characteristic fingerprint of Angong Niuhuang Pill in order to identify the Angong Niuhuang Pill containing different varieties Bovis Calculus. Methods: Water-saturated dichloromethane-methanol- acetic acid(100:50:2) was used as the extraction solvent of Angong Niuhuang Pill, then the extraction solvent was evaporated and dissolved. The extracted solution was analyzed on a C18 reversed-phase column, and eluted by acetonitrile-water (0.5% phosphoric acid) in a gradient mode. The detection of wavelength was 200nm. Results: Through the extraction solvent, detection wavelength and methodological study, a Bovis Calculus different varieties and their preparations Angong Niuhuang characteristic fingerprint were established and the characteristic peaks of natural Bovis Calculus in the fingerprint of Angong Niuhuang Pill were determined. It turned out that the characteristic peaks fingerprint was used to identify natural or cultured Bovis Calculus and Angong Niuhuang Pill containing natural or cultured Bovis Calculus, which showed the stability and reproducibility in application. Conclusion: This characteristic fingerprint method of Angong Niuhuang Pill can reflect the characteristics of natural Bovis Calculus in Angong Niuhuang pills, and identify the Angong Niuhuang Preparations containing natural Bovis Calculus from cultured Bovis Calculu, which can be used for the overall quality evaluation of Angong Niuhuang Pill.
