摘要
以带腋芽的裸花紫珠(Callicarpa nudiflora Hook.et Arn.)嫩茎段为外植体,研究其组培快繁技术.结果表明,用0.1%的升汞处理外植体15 min,接种于MS+1.5 mg/L 6-BA+30 g/L蔗糖培养基上,7d后萌发腋芽;丛生芽在MS+2.0 mg/L 6-BA+30 g/L蔗糖的培养基增殖效果最佳,增殖系数达8.50;丛生芽的继代培养周期是30 d,超出40 d时会有死株现象;培养基MS+0.5~0.8 mg/L NAA+0.5 g/L活性炭+30 g/L蔗糖适宜诱导生根,生根率达100%;生根苗移栽在V河沙∶V木屑∶V园土=1∶1∶1的混合基质中,塑料薄膜保湿20 d成活率达93%,且植株生长良好.
Using tender stem segments of Callicarpa nudiflora as explants,the techniques of tissue culture and rapid propagation in vitro were studied.Results showed that the optimum disinfector was 0.1% HgCl2 for 15 min,with MS+6-BA1.5 mg/L+ sucrose 30 g/L as the initial medium.After inoculation for 7 d,the axillaries buds began to germinate.Using MS+6BA 2.0 mg/L + sucrose 30 g/L as the proliferation medium,the proliferation multiples reached 8.50.The best cycle of subculture training was 30 d.The plantlets would appear dead when more than 40 d.The rooting culture medium was MS+ NAA 0.5~0.8 mg/L+ active carbon 0.5 g/L + sucrose 30 g/L,with the best rooting effect and the rate of rooting of 100%.When the rooted seedlings were transplanted into the mediums of sand∶soil∶sawdust (1∶1∶1) and covered with plastic film for keeping moisture for 20 d,the survival rate reached up to 93%.
