摘要
目的以马鹿鹿角盘为原料提取胶原蛋白,对其胶原蛋白的结构、分子量分布和氨基酸组成进行研究,为今后马鹿产品开发奠定基础。方法采用乙酸和胃蛋白酶结合法提取马鹿鹿角盘胶原蛋白;利用紫外扫描、红外光谱分析、SDS-PAGE垂直电泳和高效液相色谱法研究马鹿鹿角盘胶原蛋白的结构、分子量分布和氨基酸组成。结果紫外扫描分析显示,马鹿鹿角盘胶原蛋白在205 nm处有最大吸收峰,符合胶原蛋白的特性。红外光谱显示,马鹿鹿角盘胶原蛋白保留了完整的三股螺旋结构。SDS-PAGE垂直电泳显示,胶原蛋白含有α1、α2和β链3条链。氨基酸分析表明,马鹿鹿角盘胶原蛋白具有典型Ⅰ型胶原蛋白的氨基酸组成。结论从马鹿鹿角盘中提取的胶原蛋白属于Ⅰ型胶原蛋白。
Objective The collagen, extracted from the antler base of Cervus elaphus, was studied for the structure, molecular weight distribution and amino acid composition of its collagen, to lay the foundation for the future development of red deer products. Methods Collagen was extracted from antler base of Cervus elaphus using acetic acid and pepsin. The structure, molecular weight distribution and amino acid composition of collagen from antler base of Cervus elaphus were analyzed by ultraviolet spectra, infrared spectroscopy, SDS-PAGE and high performance liquid chromatography ( HPLC ) respectively. Results The maximum absorption wavelength of collagen from antler base of Cervus elaphus was 205nm within ultraviolet spectra, and was in accordance with the characteristics of collagen. Infrared spectroscopy results showed that extracted collagen comprised three different chains of α1, α2 and β. Analysis on HPLC revealed that the amino acid composition of extracted collagen from antler base of Cervus elaphus cohered with that of type I collagen typically. Conclusion Collagen extracted from antler base of Cervus elaphus belongs to type I collagen.
