蛹虫草菌产腺苷的培养基优化

摘要 以蛹虫草菌丝体中腺苷含量为指标,进行发酵培养,优化液体发酵培养基。通过单因素实验结果,选出最适宜的碳源是玉米淀粉,氮源是玉米浆粉和NH4Cl,金属离子是ZnSO4;然后采用9因素2水平Plackett-Burman设计实验得出对蛹虫草发酵水平影响显著的因素,分别为玉米淀粉、玉米浆粉和MgSO4;最后运用响应面分析方法对3个因素3个水平进行优化,获得最优组合浓度为玉米淀粉13.35g/L、玉米浆粉45.74g/L、MgSO4为0.40g/L,此时菌体腺苷含量为4 751μg/g。验证结果表明蛹虫草腺苷含量提高了210%,优化效果极为显著。 The liquid fermentation medium of Cordyceps militaris was optimized based on the adenosine content in mycelium. Firstly, through the single factor comparing experiment, maize starch was selected as the best carbon source and corn steep power and NH4Cl were the best nitrogen source, while ZnSO4 was determined as the most suitable metalion. Secondly, the results of a Plackett - Burman design showed that in 9 factors maize starch, steep power and MgSO4 were the most significant factors for were determined by response surface method (RSM). the fermentation. Thirdly, the optimal levels of these factors It was finally found that when the concentration of maize starch, steep power and MgSO4 was 13.35 g/L, 45.74 g/L and 0.396 6 g/L, respectively, the yield of adenosine reached 4 751 μg/g , increasing by 210%.
