摘要
从湖北省武汉市郊区草莓种植户的大棚内采集萎蔫的植株,分离致病菌,采用PCR扩增、病原菌形态鉴定、致病性、分子检测等方法检测草莓枯萎病菌。结果表明,形态观察为尖孢镰刀菌(Fusarium oxysporum),接种草莓后出现典型的草莓枯萎病菌症状;采用真菌的通用引物对ITS1和ITS4进行PCR检测,分离得到的致病菌株均能扩增出分子量为567 bp的特异性条带。结合形态、致病性测定和分子检测,确定分离和鉴定的病原菌为草莓枯萎病菌(Fusarium oxysporum Schlecht.f.sp.fragariae Winks et Williams)。
Wilting strawbeny in the greenhouse of strawbeny growers in the suburb of Wuhan city of Hubei province was used to explore a PCR detection technology of Fusarium oxysporum Schlecht.f.sp.fragariae Winks et Williams.DNA bands of 567bp were obtained from root,leaf,blossom and fruit isolates of inffected hosts with PCR foucusing on ITS1 and ITS4 regions by unversal fungal primers.DNA sequencing results showed that these amplified products had high homology with the related sequence in GenBank,NCBI.Combined with pathogen inoculation and isolation of root,leaf,blossom and fruit,morphological obsmvation and pathogenicity,the unversal primer-PCR established in this study could be a basic method of detecting Fusarium oxysporum Schlecht.f.sp.fragariae Winks et Williams.
