摘要
利用贵州黑山羊构建DNA池,设计引物扩增STAT5A基因第13外显子及部分内含子序列。利用生物信息学软件分析SNPs位点对STAT5A基因RNA二级结构、STAT5A蛋白二级及三级结构的影响。结果表明,在扩增的STAT5A基因中筛选到2个SNPs:T+1641G和G+1248T,其中T+1641G为脯氨酸(Pro)的同义突变,G+1248T为错义突变,导致编码的缬氨酸(Val)变为苯丙氨酸(Phe)。SNPs位点对STAT5A基因RNA二级结构和STAT5A蛋白结构均有一定影响。
Guizhou Black Goat was used to construct DNA pools.The complete sequences of the 13th coding exon and part of intron region in STA T5A were obtained with one pair of specific primers.The effects of SNPs loci on the secondary structure of RNA,secondary and tertiary structure of STA T5A protein were analyzed with bioinformatics.The results showed that two SNPs (T+1641G and G+1248T) were found in STAT5A,which included one synonymous mutations of proline (T+1641G,Pro) and a missense mutations changed the coded valine to phenyalanine (G+1248T,Val→Phe).The SNPs loci had certain effect on the RNA secondary structure of STA T5A gene and the coded protein structure.
