摘要
以解淀粉芽孢杆菌T1为研究菌种,从5种培养基中选出最有利于其生长的基础发酵培养基,并通过单因子试验和正交试验,对基础发酵培养基的配方及发酵条件进行了优化。结果表明,解淀粉芽孢杆菌T1的最优培养基配方为复合氨基酸营养液1%、蛋白胨1.5%、糖蜜5%、可溶性淀粉2.5%。最优培养条件为pH 7.0,装瓶量80 mL/250 mL、接种量5%、温度37℃、摇床转速180 r/min、发酵时间36 h。优化后的培养液中解淀粉芽孢杆菌T1活菌数由1.4×109cfu/mL增加至3.5×109cfu/mL。
Using Bacillus amyloliqueficiens T1 as strain,five well-defined culture media were selected to promote its multiplication.The proportion of each component and the culture condtions were optimized with single factor test and orthogonal test.The results showed that a maximal yield of cell amount could be obtained in the medium containing 1% compound amino acid nutrient solution,1.5% peptone,5% molasses and 2.5% soluble starch.The best cultivation conditions were pH 7.0,bottling volume 80 mL/250mL,inoculum size of 5%,temperature 37 ℃,shaking speed 180r/min,culture time 36 h.Compared with the initial conditions,the optimized medium and culture conditions resulted in a remarkable increase of the strain multiplication.The maximum yield of the bacteria cells was improved from 1.4×109 cfu/mL to 3.5×109 cfu/mL.
