南海区卵形鲳鲹遗传多样性的研究 被引量:4

摘要 为研究南海区4个不同地理位置卵形鲳鲹种群(QZ、ZJ、BH、HN)的遗传多样性。利用35对可扩增出特异性条带的引物对卵形鲳鲹4个不同地理种群进行遗传多样性分析,筛选出10对扩增效果比较好的引物,其中7对引物具有多态性。7个微卫星多态位点中,一共扩增出21个等位基因;多态信息含量为0.2896~0.6914,且处于中度到高度多态,表明这些引物可作为卵形鲳鲹遗传多样性分析的有效微卫星引物;4个群体的平均期望杂合度在0.4811~0.6135,北海群体的平均期望杂合度小于0.5,表明北海卵形鲳鲹群体的遗传多样性较低。用Popgene32软件计算出4个种群卵形鲳鲹的标准遗传距离和相似性系数,并构建聚类树,发现湛江卵形鲳鲹群体和海南群体遗传相似性系数最大,遗传关系最近。 The genetic diversity of 4 different Trachinotus ovatus groups in Nanhai district ( QZ, ZJ, BH,HN) was analyzed by using the selected micro satellite sequences. 35 pairs of primers were amplified successfully and every primer could produce a single, bright band, which were used to analyze the genetic diversity of 4 different Trach/notus ovatus groups in Nanhai district. The result that the amplification effect of 10 pairs of polymorphic primers was batter, in which 7 pairs of primers expressed polymorphic, we amplified 21 alleles in 4 groups. In 4 dif- ferent Trachinotus ovatus groups, the number of the effective allele was 1 -3. 8413, on the average of 2.40; the information content (PIC) was 0. 2896 -0. 6914, the result indicated that these molecular markers could be used to analyzed the genetic diversity of Trach/notus ovatus; the mean expected hcteI^zygesis wad 0.4811 -0.6135, and then the mean expected heterezygosis of BeiHai group was less than 0.5, indicating that the genetic diversity of the group was low. The genetic identity and genetic distance among 4 gToups were estimated by the Popgene32 software. The results showed that the genetic distance between ZJ and HN was near,and their genetic similarity were high.
