摘要
目的观察小柴胡汤对肝纤维化大鼠肝组织细胞色素P450 2E1(cytochrome P450 2E1,CYP2E1)表达的影响。方法皮下注射10%CCl45 mL/kg,2次/周,连续14周,制备肝纤维化模型。实验分为空白对照组,模型组,小柴胡汤低剂量组(1g/kg)、中剂量组(2g/kg)、高剂量组(4g/kg),小柴胡汤从制模第9周开始灌胃给药。第14周末,形态学行HE染色观察小柴胡汤对肝纤维化程度的影响;免疫组化观察CYP2E1蛋白表达水平;Real-time RT-PCR检测肝纤维化大鼠肝脏CYP2E1 mRNA的表达。结果与模型组相比,小柴胡汤各剂量组肝纤维化病理改变均有不同程度的恢复;与空白对照组比较,模型组肝组织CYP2E1蛋白及mRNA表达明显升高(P<0.05);小柴胡汤各剂量组较模型组在一定程度下调CYP2E1蛋白及mRNA的表达。结论小柴胡汤可治疗CCl4引起的大鼠肝纤维化并下调肝组织CYP2E1的表达。
Objective To observe the effect of Xiao -Chai -Hu Tang on CYP2E1 expression in rats with liver fibrosis. Methods Liver fibrosis model was prepared by injecting 10% CCl4, 5 mL/kg, twice a week for consecutive 14 weeks. Rats were divided into control group, model group, low dose group( lg/kg), middle dose group (2g/kg), and high dose group(4g/kg) of Xiao - Chai - Hu Tang. At the end of the 14th week, the effect of Xiao - Chai - Hu Tang on liver fibrosis was observed by HE staining. The level of CYP2E1 protein was observed by immunohistochemistry. Real - time RT - PCR was used to detect the expression of CYP2E1 mRNA. Results Compared with model group,liver fibrosis pathology was recovered in each dose group of Xiao - Chai - Hu Tang. CYP2EI protein and mRNA expression was significantly increased in model group compared with normal group (P 〈0.05 ). Each dose group of Xiao Chaihu Tang, compared with model group, decreased CYP2E1 protein and mRNA expression. Conclusion Xiao - Chai - Hu Tang had a therapeutic effect on rat liver fibrosis induced by CC14and reduced the over -expression of CYP2E1 in the liver.
