藏红花提取液对慢性高眼压兔视网膜的影响 被引量:4

摘要 目的建立兔慢性高眼压模型,分析藏红花提取液对慢性高眼压兔眼视网膜的作用效果。方法选取120只新西兰家兔,随机分为对照组、高眼压模型组以及干预治疗组,每组40只共80眼。高眼压模型组以及干预治疗组家兔通过注射复方卡波姆溶液建立慢性高眼压模型,干预治疗组家兔每日静脉推注藏红花提取液。在建立模型后的7 d、14 d以及4周,各组分别处死10只动物,利用光镜观察视网膜形态学变化,并检测视网膜超氧化物歧化酶(SOD)、丙二醛(MDA)以及视网膜神经节细胞(RGCs)水平,对结果进行统计学分析。结果高眼压模型组及干预治疗组视网膜均发生肿胀、萎缩,与高眼压模型组相比,干预治疗组损伤程度较轻;建立模型7d时,高眼压模型组及干预治疗组SOD值明显低于对照组,MDA含量显著高于对照组,差异有统计学意义(P<0.05),但干预治疗组指标变化幅度明显小于高眼压模型组(P<0.05);建立模型14 d时,高眼压模型组及干预治疗组SOD明显恢复,接近正常水平,MDA含量仍高于对照组,差异有统计学意义(P<0.05);干预治疗组RGCs数目为(287.62±23.54)个,与对照组比较差异无统计学意义(P>0.05),高眼压模型组RGCs数目为(62.78±10.24)个,显著低于对照组及干预治疗组,差异有统计学意义(P<0.05)。结论藏红花提取液能够保护慢性高眼压兔眼视网膜功能,可作为青光眼的有效治疗药物。 Objective To establish a rabbit model of chronic ocular hypertension, analyze saffron extract effects for retina of rabbits with chronic high intraocular pressure. Methods 120 New Zealand rabbits were randomly divided into control group, model group and high IOP intervention group, 40 cases (80 eyes) in each group. Ocular model group and in-tervention group were established rabbits chronic ocular hypertension model by injecting carbomer solution, rabbits in the intervention group were given daily intravenous injection of saffron extract. After model establishment for 7 d, 14 d and 4 week, each group were sacrificed 10 animals. Retinal morphological changes were observed by light microscope, then superoxide dismutase (SOD), malondialdehyde (MDA), retinal ganglion cells (RGCs) levels were detected. The re-sults were analyzed. Results The retinal had swelling, shrinking in the intervention group and model group, compared with the model group, the degree of damage was less in the intervention group. After model establishment for 7 d, SOD levels in the model group and intervention group were significant lower than the control group, and MDA levels in the model group and intervention group were significant higher than the control group, with statistically significant differ-ence (P〈0.05), but the rangeability in the intervention group was less than the model group (P〈0.05). After model es-tablishment for 14 d, SOD levels in the model group and intervention group had recovered to normal level, but MDA levels still higher than the control group, with statistically significant difference (P〈 0.05). The number of RGCs in the intervention group was (287.62±23.54), compared with the control group, the difference was not statistically significant (P〉0.05). The number of RGCs in the model group was (62.78±10.24), which was significantly lower than the interven-tion group and the control group, with statistically significant difference (P〈0.05). Conclusion Saffron extract can pro-tect retinal function of rabbits with chronic high intraocular pressure, can be used as an effective therapy for glaucoma.
