摘要
目的:探讨Survivin、Caspase-3、Bcl-2和Bax在宫颈鳞癌患者血液、癌组织中的表达及其相互关系。方法:对123例高危患者(30例宫颈炎、41例宫颈内瘤变、52例宫颈癌)抽取血液,采用ELISA方法检测血清中Survivin、Caspase-3、Bcl-2和Bax蛋白的含量,采用流式细胞术检测血细胞中Survivin、Caspase-3、Bcl-2和Bax的阳性率并结合临床病理特点分析。结果:宫颈炎患者血清中Survivin、Bcl-2蛋白含量及血细胞中Survivin、Bcl-2阳性率明显低于宫颈内瘤变和宫颈癌患者,差异有统计学意义(P<0.01);且宫颈内瘤变患者明显低于宫颈癌患者,差异有统计学意义(P<0.01)。宫颈炎患者血清中Caspase-3、Bax蛋白含量及血细胞中Caspase-3、Bax的阳性率明显高于宫颈内瘤变和宫颈癌患者,差异有统计学意义(P<0.01);且宫颈内瘤变患者明显高于宫颈癌患者,差异有统计学意义(P<0.01)。结论:宫颈癌患者Survivin、Bcl-2蛋白的上调和Caspase-3、Bax蛋白的下调在宫颈癌发生、发展过程中发挥着即独立又协同的作用。
Objective: To explore the expressions of Survivin, Caspase -3, Bcl -2 and Bax in blood and cancer tissue of patients with cervical squamous cell carcinoma and their interrelation. Methods: Blood samples were abstracted from 123 high risk patients; 30 patients with cervicitis, 41 patients with cervical intraepithelial neoplasia (CIN) and 52 patients with cervical cancer were included, ELISA was used to detect the serum contents of Survivin, Caspase - 3, Bcl - 2 and Bax proteins ; flow cytometry was used to detect the positive rates of Survivin, Caspase - 3, Bcl - 2 and Bax in blood cells, the clinicopathological characteristics were analyzed. Results : The serum contents of Survivin, Bcl - 2 proteins and the positive rates of Survivin, Bcl - 2 in blood cells of patients with cervicitis were statistically significantly lower than those in patients with CIN and cervical cancer (P 〈 0. 01 ) ; the serum contents of Survivin, Bcl -2 proteins and the positive rates of Survivin, Bcl - 2 in blood cells of patients with CIN were statistically significantly lower than those in patients with cervical cancer ( P 〈 0.01 ) ; the serum contents of Caspase - 3, Bax proteins and the positive rates of Caspase - 3, Bax in blood cells of patients with cervicitis were statistically significantly higher than those in patients with CIN and cervical cancer ( P 〈 0. 01 ) ; the serum contents of Caspase - 3, Bax proteins and the positive rates of Caspase - 3, Bax in blood cells of patients with CIN were statistically significantly higher than those in patients with cervical cancer (P 〈 0. 01 ) .Conelusion: Upregulation of Survivin, Bcl -2 proteins and Downregulation of Caspase-3, Bax proteins in cervical cancer patients play independent and synergistic effects in occurrence and development of cervical cancer.
