摘要
以PCR技术及琼脂糖凝胶电泳技术为基础,对转基因水稻克螟稻2号和阴性受体秀水11进行真实性鉴定。利用典型性检测、指纹图谱技术对候选物的转基因事件的真实性、转入事件正确性(是否为特异性目标外源基因)、品系遗传背景进行鉴定,结果表明,克螟稻2号只含特异性序列Cry1Ab,秀水11不含所选的外源基因,它们具有相同的遗传背景。随机各抽取克螟稻2号和秀水11水稻100株,通过特异性PCR、免疫印迹实验来检测原材料的纯度,结果表明,克螟稻2号为100%的转基因水稻,秀水11为100%的阴性受体水稻。
Based on PCR and agarose gel electrophoresis, the authenticity of identification of transgenic rice KMD 2 and negative receptor Xiushui 11 is researched. The detection of special transgenic gene and other foreign genes in raw materials is by typicality identification. No other foreign genes are detected except special foreign gene Cry1Ab in transgenic rice KMD 2, and no foreign genes is detected in Xiushui 11. Based on fingerprint technology, including Xiushui 11 and KMD 2 there are 20 rice varieties, and the results showed that Xiushui 11 and KMD 2 have the same fingerprint and also be different from other 18 rice varieties. Random sampling for KMD 2 and Xiushui 11, and take special PCR and immunoblotting technique for the selected samples purity, KMD 2 are 100% transgenic rice and Xiushui 11 are 100% negative receptors rice.
