摘要
优化板蓝根颗粒中多糖的提取,并测定其单糖组成及含量。采用正交试验设计优化板蓝根颗粒中多糖的提取条件。所提取多糖采用1 mol/L硫酸95℃恒温降解5 h,以1-苯基-3-甲基-5-吡唑啉酮(PMP)作为单糖的柱前衍生化试剂,采用毛细管区带电泳法测定。结果显示,板蓝根颗粒中的多糖由木糖、阿拉伯糖、葡萄糖、鼠李糖、甘露糖、半乳糖、葡萄糖醛酸和半乳糖醛酸等组成,其中葡萄糖醛酸和半乳糖醛酸尚未见报道。
The extraction conditions of the polysaccharides from granules of Radix Isatidis were optimized using orthogonal design, and the monosaccharide composition of the polysaccharide was determined. The polysaccharides were completely hydrolyzed into monosaccharides with 1 mol/L sulfuric acid at 95℃ for 5 h. Then, the monosaccharides were derivatized by 1-phenyl-3-methyl-5-pyrazolone (PMP) and were determined by capillary zone electrophoresis. The results indicated that the polysaccharides of granules of Radix Isatidis were composed of xylose, arabinose, glucose, rhamnose, galactose, mannose, glucuronic acid and galacturonic acid, in which glucuronic acid and galacturonic acid were not reported preveniently.
