摘要
研究荭草花提取物中原儿茶酸、异荭草素、荭草素、牡荆素、木犀草苷、花旗松素、山柰素-3-O-α-D-葡萄糖苷、槲皮苷和山柰素-3-O-β-L-鼠李糖苷9种主要有效成分在大鼠小肠的吸收特征。采用外翻肠囊法进行离体肠吸收试验,以UPLC-MS/MS法测定各成分含量,分析各成分在不同部位的吸收特性。荭草花提取物中上述9种成分在不同肠段的吸收均呈线性吸收,且符合零级吸收速率,肠吸收速率常数(Ka)随提取物给药剂量的增加而增加,表明其为被动吸收,各成分在各肠道的吸收趋势均为十二指肠、空肠、回肠优于结肠。
The absorption characteristics of the main active components in extract of the flowers of Polygonum orientale, including protocatechuic acid, isoorientin, orientin, vitexin, galuteolin, texifolin, kaempferol-3-O-β-D- glucoside, quercitrin and kaempferol-3-O-α-L-rhamnoside in rats' small intestine were investigated. Experiments were conducted with the help of the rat everted gut sacs model. The concentrations of all compositions were determined by UPLC-MS/MS, and the absorption at different intestinal segments was observed. The absorption of each component was linear in different intestinal segments at different doses, and it was consistent with zero order rate process. The absorption rate constant (K.) of nine constituents increased with the increasement of the extract concentration, which was consistent with passive absorption. The absorption of nine components in duodenum, jejunum and ileum was greater than in the colon.
