摘要
建立了高效液相色谱法测定新型靶向抗肿瘤化合物SKLB710与大鼠血浆蛋白的结合率。采用C18色谱柱,流动相为水∶甲醇(30∶70),检测波长268 nm。应用平衡透析法结合HPLC-UV法测得高、中、低浓度SKLB710与大鼠血浆蛋白的结合率分别为(95.8±0.2)%、(95.4±1.0)%和(96.2±0.5)%。
An HPLC method was established for the determination of the rat plasma protein binding rate of targeted anticancer lead compound SKLB710. A C18 chromatographic column was used, with the mobile phase of water : methanol (30 : 70) at the detection wavelength of 268 nm. The rat plasma protein binding rate of SKLB710 was determined by equilibrium dialysis and HPLC method. The plasma protein binding rates of SKLB710 at high, medium and low concentrations were (95.8±0.2) %. (95.4±1.0) % and (96.2±0.5) %.
