硬脂醇半乳糖苷修饰盐酸吉西他滨纳米脂质体的包封率测定 被引量:2

摘要 建立了微柱离心-高效毛细管电泳法测定硬脂醇半乳糖苷(18-Gal)修饰的盐酸吉西他滨纳米脂质体包封率。采用阳离子交换树脂分离脂质体和游离药物。采用未涂渍标准熔融石英毛细管柱,以pH 2.69的45 mmol/L磷酸二氢钠溶液为电泳缓冲液,运行电压25 kV,毛细管柱温25℃,检测波长269 nm,压力进样5 kPa×3 s。盐酸吉西他滨在4∽250?g/ml浓度范围内线性关系良好。3批制品包封率分别为70.9%、69.7%和72.2%。 A microcolumn centrifugation-HPCE method was established for the determination of the entrapment efficiency of stearyl alcohol galactosidase (18-Gal) modified gemcitabine hydrochloride nanoliposomes. The cation exchange resin method was employed to separate the free drug from the liposomes. A standard uncoated fused silica capillary column was used with the electrophoresis buffer of 45 mmol/L sodium dihydrogen phosphate solution (adjusted to pH 2.69 with phosphoric acid). The separation voltage was 25 kV, column temperature was 25 ~C, detection wavelength was 269 nm, the pressure injection was 5 kPax3 s. The calibration curve was linear in the concentration range of 4 - 250 ~tg/ml. The average entrapment efficiencies of three batches of liposomes were 70.9 %, 69.7 % and 72.2 %, respectively.
