王不留行中总黄酮和总皂苷的分离及活性研究 被引量:9

Separation and active study of the flavonoids and total saponins from Vaccaria segetalis

摘要 目的分离王不留行中的总黄酮和总皂苷,并分别评价其对人微血管内皮细胞(HMEC-1)增殖的影响。方法采用大孔树脂一步法对王不留行活性物质进行分离,通过理化分析对各类活性物质进行鉴别,采用SRB法评价各组分对HMEC-1增殖的影响。结果 30%乙醇洗脱组分为王不留行的总黄酮,70%乙醇洗脱组分为王不留行总皂苷;分离得到的王不留行总黄酮有促进HMEC-1细胞增殖的活性,王不留行总皂苷对HMEC-1的增殖表现出较好的抑制活性,其IC50值达1.68μg·mL-1。结论王不留行总黄酮和总皂苷对HMEC-1细胞的增殖表现出相反的作用。 OBJECTIVE To separate the flavonoids and total saponins from Vaccaria segetalis and investigate the effect to human microvascular endothelial cells ( HMEC - 1 ) , respectively. METHODS By macroporous resin, the active substances in Vaccaria segetalis were isolated in one step, then physical and chemical analysis are carried out to identify the active substances isolated by macroporous resin, finally, the effect of each component to HMEC - 1 was evaluated by SRB assay. RESULTS The 30% and the 70% ethanol eluted fractions of HPD - 100 macroporous resin were flavonoids and total saponins from Vaccaria segetalis, respectively. The ilavonoids of Vaccaria segetalis exhibited a promotion activity to HMEC - 1 cell proliferation, however, the total saponins of Vaccaria segetalis exhibited an inhibition activity to HMEC - 1 cell proliferation and the IC_50 reached 1.68 μg·mL^-1. CONCLUSION The flavonoids and total saponins from Vaccaria segetalis exhibited an opposite activity towards HMEC - 1 cell proliferation.
