摘要
目的观察白藜芦醇对妊娠糖尿病大鼠血糖的作用及机制。方法采用高脂膳食诱导雌性大鼠6周后,按照1∶2雄雌比例合笼过夜,阴道涂片镜检见精子者记为妊娠0 d,选妊娠1 d的大鼠腹腔注射35 mg/kg链脲佐菌素诱导糖尿病模型。正常妊娠大鼠为对照组,随机将30只妊娠糖尿病大鼠按血糖水平随机分为3组(n=10):妊娠糖尿病模型组(GDM)、白藜芦醇低、高剂量组。干预14 d后观察血清血糖、胰岛素、血脂及炎症因子的变化。结果与对照组相比,妊娠糖尿病模型大鼠空腹血糖、胆固醇(TC)、甘油三酯(TG)、游离脂肪酸(FFA)、肿瘤坏死因子α(TNF-α)和C反应蛋白(CRP)显著升高,胰岛素和HDL-C显著降低;与GDM组相比,白藜芦醇能够显著降低妊娠糖尿病大鼠的血糖TG、FFA、TNF-α和CRP的水平,升高胰岛素水平,但对TC和HDL-C无影响。结论白藜芦醇具有降低妊娠期糖尿病大鼠血糖和改善胰岛素抵抗的作用,其机制可能是通过改善脂代谢和炎症来实现的。
Objective To investigate the treatment effect of resveratrol on blood glucose in gestational diabetes mellitus and its underlying mechanism. Methods All rats were induced by high fat diets for 6 weeks,then female and male rats were caged together with the proportion of 1∶ 2. The time when sperm cells were found in female rats under microscope was recorded as pregnancy 0d. Rats with gestation for 1 day were injected with 35 mg /kg of STZ once through abdominal cavity. Normal rats served as control group. Thirty model rats were randomized into GDM group,the low and high doses of resveratrol groups. Fasting serum glucose,insulin,blood lipids and inflammatory factors were observed after intervention for 14 days. Results Compared to controls,fasting serum glucose,cholesterol( TC),triglycerides( TG),free fatty acids( FFA),tumor necrosis factor α( TNF-α) and C-reactive protein( CRP) were significantly increased,but serum insulin and high density lipoprotein cholesterol( HDL-C) in gestational diabetic rats. Compared to gestational diabetic rats,fasting serum glucose,TG,FFA,TNF-α and CRP were significantly decreased and serum insulin was significantly increased in the resveratrol intervention groups. However,there was no difference in TC and HDL-C levels between gestational diabetes mellitus group and the resveratrol intervention groups. Conclusion Resveratrol can decrease the serum glucose and improve insulin resistance in gestational diabetes mellitus rats,the mechanism impossibly involving the improving inflammation and the lipids metabolism.
