重力矢量及趋化诱导对脂肪干细胞在PLGA电纺膜上早期增殖的影响 被引量:2

摘要 观察研究重力矢量方向及血小板衍生生长因子(PDGF)趋化诱导,在体外环境对人脂肪于细胞(ASCs)在PLGA电纺膜表面黏附和增殖活动的影响.体外原代培养从人颊脂垫提取的ASCs,根据不同重力矢量条件以及有无趋化因子PDGF将实验分为4个组.A组:PDGF/重力矢量(+),含PDGF的PLGA电纺膜置于ASCs悬液底部;B组:PBS/重力矢量(+),空白(PBS)电纺膜置于ASCs悬液底部;C组:PDGF/重力矢量(-),含PDGF的PLGA电纺膜覆于ASCs悬液表面;D组:PBS/重力矢量(-),空白(PBS)电纺膜覆于ASCs悬液表面.分别于0、1、3、5、8、11d加入Alamar Blue继续孵化24 h后测OD值,对照标准曲线,计算各组Alamar Blue的下降百分比,评估ASCs的黏附增殖活动.结果显示:在重力矢量(+)组的早期(第1~4 d),无论趋化因子PDGF存在与否,Alamar Blue的下降百分比都能达到30%左右,不影响细胞的定植、黏附与增殖.但是在重力矢量(-)组,缺少趋化因子PDGF的Alamar Blue的下降百分比为25%左右,细胞将无法正常的定植与黏附.PDGF具有明显诱导ASCs趋化聚集作用,能减轻不利重力因素的影响,显著改善ASCs黏附与增殖.
