模拟失重条件下大鼠心室肌Beclin-1和LC3的表达变化 被引量:2

摘要 目的探讨模拟失重大鼠心室肌自噬相关基因Beclin-1和微管相关蛋白轻链3(LC3)的表达变化。方法雄性SD大鼠12只,随机分为对照组和模拟失重组。模拟失重4周后取心室肌组织,采用RT-PCR和Western Blot方法测定Beclin-1和LC3 mRNA及蛋白表达的变化。结果与对照组相比,模拟失重4周后大鼠心室肌Beclin-1和LC3 mRNA及蛋白表达均显著增强(P<0.05)。结论模拟失重可诱导大鼠心室肌自噬相关基因Beclin-1和LC3的高表达,提示模拟失重可能引起了心室肌自噬的激活。 Objective To explore the expression of autophagy related gene Beclin-1 and LC3 in the ventricular muscle after simulated weightlessness. Methods A total of 12 male SD rats were randomly assigned to control and simulated weightlessness groups. The ventricular muscle tissues of rats subjected to simulated weightlessness were taken after 4 weeks of simulated weightlessness,respectively. RT-PCR and Western Blot were applied to determine the expression of autophagy related gene Beclin-1 and LC3. Results Compared with the control group,the expression of Beclin-1 and LC3 mRNA and protein in the ventricular muscle were both significantly increased after 4 weeks of simulated weightlessness. Conclusion Simulated weightlessness increases the expression of autophagy related gene Beclin-1 and LC3,suggesting autophagy may be induced by simulated weightlessness in the ventricular muscle of rats.
