Tissue Culture and Rapid Propagation of Haworthia cymbiformis

摘要 以多肉植物水晶掌叶片为外植体,采用MS基本培养基,附加不同的植物生长调节剂组合,研究了水晶掌愈伤组织的器官发生和植株的再生技术.实验结果表明:A1(MS+BA 3.0+NAA 0.3+3%蔗糖+0.7%琼脂)配方最为适合愈伤组织的诱导;B4(MS+BA 4.0+NAA 0.2+3%蔗糖+0.7%琼脂)配方最为适合愈伤组织的增殖;C1(MS+BA 3.0+NAA 0.2+3%蔗糖+0.7%琼脂)最为适合愈伤组织分化,芽分化较快,苗健壮,并且有丛生芽;促进生根的最佳配方为D4(1/2 MS+IBA 1.0+3%蔗糖+0.7%琼脂);组培苗经炼苗后,移栽成活率在95%以上,以此建立水晶掌试管苗无性繁殖体系. The organogenesis and plant regeneration of callus of Haworthia cymbiformis was studied by using its leaves as initial explants, MS basic culture medium and adding different plant growth regulators. The test results were as follows. MS + BA 3.0 + NAA 0. 3 + 3% sucrose + 0. 7% agar (formula A1 ) was most suitable for callus induction. MS + BA 4.0 + NAA 0.2 +3% sucrose +0. 7% agar (formula B4) was most suitable for callus proliferation. MS + BA 3.0 + NAA O. 2 + 3% sucrose + 0. 7% agar ( formula C1 ) was the best medium of buds differentiation for faster differentiation, robust seedlings, and more buds. The best formula for root was 1/2 MS + IBA 1.0 +3% sucrose +0.7% agar (formula 134). The survival rate of transplanting of culture seedlings (after being hardened) was as high as 95%. A test-tube plantlet cloning of Haworthia cymbiformis was established.
