人与十种寄生虫HMGB1序列比对分析

摘要 目的:确定人与寄生虫高迁移率族蛋白B1(HMGB1)的相似性关系。方法:运用分子生物学工具软件Generous检索NCBI数据库HMGBl氨基酸序列,比对分析寄生原虫、寄生丝虫和寄生吸虫的序列相似性以及氨基酸组成,比对分析人HMGB1炎症相关序列与10种寄生虫HMGB1的相似性。结果:3种寄生原虫、3种丝虫和4种吸虫的HMGBl序列位点一致性分别为69.2%、93.1%、68.3%;人HMGB1炎症相关序列与寄生虫HMGBl序列位点一致性50.7%。结论:人与寄生虫HMGBl序列同源,炎症相关序列与寄生虫HMGBl序列一致,人与寄生虫HMGBl促炎症作用可能存在构象和通路差异。 Objective:To determine the similarity between the sequences of the high mobility group protein B1(HMGB1)in human and parasites.Methods:Sequence alignment of HMGBl in the NCBI database was conducted by the Generous software.Sequence similarity and amino acid composition of HMGBl in the parasitic protozoa,trematode and filarial were analyzed,and the sequence similarity of human HMGB1 sequence associated with inflammation and that of ten species of parasite were analyzed and compared.Results:In three species of parasitic protozoa,three species of parasitic filarial and four species of parasitic fluke,the rate of identical sites of HMGBl was respectively 69.2%,93.1% and 68.3%.In human HMGB1 sequence associated with inflammation and parasite HMGBl sequence,the rate of identical sites of HMGBl was50.7%.Conclusion:The human and parasite HMGBl sequences were homologous and the human HMGBl sequence associated with inflammation and parasite HMGBl sequence were similar.The pro-inflammatory effects of human HMGBl and parasite HMGBl may exist differences in conformation and signal pathways.
