摘要
目的建立地埝药材中没食子酸的RP-HPLC测定方法。方法采用高效液相色谱法,色谱柱:Cosmosil C18(4.6mm×250mm,5μm);流动相为甲醇-0.5%磷酸溶液;检测波长为274nm;流速为1.0 mL·min^-1;柱温为25℃。结果没食子酸在0.234~1.794μg内线性关系良好(r=0.9999),平均回收率为99.33%,RSD=1.19%。结论该方法准确、简便、重现性好,适用于没食子酸的含量测定,有助于完善地埝药材的质量控制方法。
Objective To establish a method for the determination of gallic acid in Melastoma dodecandrum Lour. from different sources by RP-HPLC. Methods HPLC method was validated on Cosmosil C_18(4.6 mm × 250 mm, 5 μm) with a mobile phase of methanol -0.5 % phosphoric acid at a flow rate of lmL/min and a column temperature of 25 ℃. The detection wavelength was set at 274 nm. Results Gallic acid in the range of 0.234~1.794 μg(r=0.9999) had a good linear relationship, the average recovery was 99.33 %, and RSD=1.19 %. Conclusion The established method is simple, feasible and reproducible, abd it can be used for the quality control of Melastoma dodecandrum Lour.
