摘要 人类白细胞抗原-G( human leukocyte antigen G , HLA-G)属于非经典的HLAⅠ类分子,其基因多态性、组织分布、结构及生物学功能等方面与经典的HLAⅠ类抗原显著不同。生理情况下, HLA-G抗原主要表达在绒毛外滋养层细胞、成人胸腺髓质、角膜等组织细胞。病理情况下, HLA-G能在多种肿瘤、器官移植物、炎性疾病和病毒感染等组织细胞中诱导性表达[1]。 HLA-G通过与免疫细胞上的抑制性受体( KIR2DL4、ILT2、ILT4)相互作用直接发挥免疫抑制功能,如抑制NK细胞和CTL介导的细胞杀伤活性,抑制B 细胞分化和DC 细胞成熟等。此外, HLA-G可诱导产生多种调节性细胞( Treg、Tr1细胞、DC-10等)及细胞因子( IL-10、TNF-α、IFN-γ等)发挥长效免疫抑制效应[2]。本文就HLA-G在免疫耐受机制中的研究进展作一综述。
