摘要
目的探讨基质金属蛋白酶-9(MMP-9)在偏头痛大鼠模型中的表达情况。方法 48只雄性SD大鼠随机分为正常对照组、阴性对照组和模型组,每组16只。正常对照组不作处理,模型组采用皮下注射硝酸甘油法制作大鼠偏头痛模型,阴性对照组注射生理盐水。应用免疫组化技术及Western blot,分别观察各组大鼠硬脑膜、三叉神经节及三叉神经颈复合体MMP-9蛋白的表达情况。结果 MMP-9在各组大鼠中均有不同程度表达,模型组中MMP-9表达明显高于阴性对照组及正常对照组(P<0.05)。结论 MMP-9在偏头痛大鼠模型中表达增强,其可能在偏头痛发病机制中发挥重要的作用。
Objective To investigate the expression of matrix metalloproteinases-9 (MMP-9) in rat model of migraine. Methods Fourty-eight male Spragne-Dawley rats were randomly and equally divided into normal control group, negative control group and model group. The rat model of migraine was .established by subcutaneous injection of nitroglycerin in model group, and saline was injected in negative control group. The rats were not treated in normal control group. The expression of MMP-9 protein in the rat dura mater, trigeminal ganglion and trigeminocervical complex was observed by using immunohistochemistry and Western blot. Results The expression of MMP-9 to some degree was detected in each group. The expression of MMP-9 in model group was significantly higher than that in the negative control group and normal 'control group (P 〈 0.05). Condusion There was strong expression of MMP-9 in rat model of migraine. MMP-9 may play an important role in the pathogenesis of migraine.
